We are seeking a highly motivated Advanced Research Assistant to join an interdisciplinary team working on a cutting-edge CRISPR-based genome editing project at the Wellcome Sanger Institute. In collaboration with MRC, the project aims to generate proof-of-concept data for a novel gene therapy platform to treat Beta Thalassemia. Unlike traditional gene therapies that correct individual mutations, our patented technology enables one-step replacement of the entire defective gene locus with a wild-type version. This approach offers a scalable solution that has the potential to transform the gene therapy landscape.

We are looking to recruit two fixed-term positions as Advanced Research Assistants for up to 9 months (until 30 June 2026). For internal applicants, this fixed-term role is available as a secondment opportunity. The postholders will work under the supervision of Dr Valentina Migliori, in the Cellular and Gene Editing Research group led by Andrew Bassett and in collaboration with MRC leveraging the Wellcome Sanger Institute's world-class expertise in genome engineering, iPSC biology, and sequencing technologies.

About The Role

You will work independently within a collaborative team of Technical Specialists, Postdocs and Staff Scientists. You will use human induced pluripotent stem cells (hiPSCs) to optimise the editing system and evaluate its efficiency and specificity using techniques such as flow cytometry (FACS), advanced long-read sequencing, ddPCR, and drug-based selection. The goal is to generate robust, high-quality data to support the potential launch of a biotech spin-out company that could provide opportunities for further career development. If successful, this platform will be expanded to additional genetic loci and diseases.

Your Key Responsibilities Will Include

  • To develop and perform CRISPR-based genome editing experiments to determine efficiency and side effects of whole gene replacement in hIPSCs, using FACS and long read sequencing.
  • To analyse the read out of long read sequencing experiments to better understand off target effects.
  • To clone and optimise reagents to target the specific genomic sites.
  • To design, perform and interpret experiments, developing the necessary practical skills where required, and to accurately document your progress both in written reports and verbally.
  • To drive project progress through contributions to lab work, and coordination with other team members and collaborating groups.
  • To contribute towards writing up research results for publication, and deal with all aspects of the publication process.
  • To participate and present progress reports at regular meetings within the lab, and communicate research by poster and oral presentations at local and international meetings.
  • To keep up to date with recent technological advancements in genome engineering (e.g. CRISPR), stem cells, gene therapy and blood disorder through the scientific literature and communicating directly with other groups.
  • To support the general duties of the wider team by participating in routine lab maintenance such as ordering of resources, organising repairs, making up solutions and dealing with suppliers to facilitate smooth running of the laboratory. Passing on skills and knowledge to other team members and visitors.

About You

You will have hands-on experience in cell culture and molecular biology, ideally with prior exposure to genome editing technologies such as CRISPR/Cas9. Experience working with human induced pluripotent stem cells (hiPSCs) or other primary cell systems will be highly advantageous. You are curious, detail-oriented, and motivated by the potential to translate cutting-edge science into impactful therapies. You enjoy working in a fast-paced, collaborative environment and can manage multiple priorities effectively. Strong organisational, communication and record-keeping skills are essential, as is a willingness to learn and adapt new experimental approaches.

About Us

We are the Cellular and Gene Editing Research team within Scientific Operations at the Wellcome Sanger Institute, led by Dr Andrew Bassett. Our team specialises in developing and applying cutting-edge technologies in genome engineering, single-cell analysis, and spatial biology to enable high-impact research. While we work across a diverse portfolio of projects, this particular role is embedded within a new translational initiative focused on pioneering genome engineering technologies for therapeutic purposes. As part of our collaborative and innovation-driven environment, you will work closely with both internal and external partners to generate proof-of-concept data for whole gene replacement strategies in human stem cells.

Knowledge, Skills, And Experience Required

  • MSc or BSc in biological sciences with relevant laboratory experience.
  • Extensive experience culturing mammalian cell lines and/or stem cells.
  • Ability to design and troubleshoot experiments independently.
  • Interest in translational research, particularly in the context of beta thalassemia.
  • Familiarity with CRISPR-based genome engineering techniques.
  • Proficiency in molecular biology techniques (e.g. cloning nucleic acid purification, sequencing library construction) and FACS analysis.
  • Excellent organisational skills, with the ability to work to timelines and deliver on project milestones.
  • Self-motivated, collaborative, and capable of working independently once trained.
  • Strong oral and written communication skills.
